G protein coupled receptors (GPCR) regulate a diverse array of intracellular signaling. cascades in response to hormones, neurotransmitters, ions, photons, odorants and other stimuli. As such, they play an essential role in physiology and disease and represent attractive drug targets.

G proteins are specialized proteins with the ability to bind the nucleotides guanosine triphosphate (GTP) and guanosine diphosphate (GDP). There are two models for GPCR-G Protein interactions: 1) ligand-GPCR binding first, then binding to G Proteins; 2) "Pre-coupling" of GPCRs and G Proteins before ligand binding (review Oldham WM and Hamm HE, 2008).
